New Features?

Yes.

(1.) Ability to queue building production.

(2.) Better pathing for air units.

(3.) Handi-cap ability for playing higher-level players on battle.net

(4.) The ability to build unique user-built shops for heros.

(5.) New creeps, tile sets, and nuetral hero mercenaries.

New units?

Yes. From what I can tell Mountain Giant(NE), Troll Batrider(ORC), Spell Breaker(HMN), Dragon Hawk or Wind Serpent(HMN). More will probably be released soon.

New Heros? If so, how many per race?

Yes. About 1 new hero per race. I recall the Blood Mage(HMN), Shadow Hunter(ORC), Crypt Lord(UD), and Assassin(NE)

Any new items?

Most likely. I believe there are going to be 15-20+ new items and summonables.

What is the plot?
